General description

Leucine-rich repeats and immunoglobulin-like domains protein 1 (UniProt: Q96JA1; also known as LIG-1) is encoded by the LRIG1 (also known as LIG1) gene (Gene ID: 26018) in human. LIG-1 is a widely expressed single-pass type I membrane protein that acts as a feedback negative regulator of signaling by receptor tyrosine kinases, through a mechanism that involves enhancement of receptor ubiquitination and accelerated intracellular degradation. LIG-1 contains 3 Ig-like C2-type (immunoglobulin-like) domains, 15 LRR (leucine-rich) repeats, 1 1 LRRCT domain, and 1LRRNT domain. The LRRs and the Ig-domains are each sufficient for EGFR/ERBB1 binding and this interaction is abolished only when these two domains are deleted. LIG-1 is highly expressed by intestinal stem cells and controls the size of the intestinal stem-cell niche by regulating the amplitude of growth-factor signaling. It is also reported to control stem-cell proliferation in epidermis. LIG-1 is reported to be expressed in a gradient with highest levels in intestinal stem cells and it is absent from Paneth cells. About one-third of all cells in the intestinal crypts are shown to express LIG-1. (Ref.: Wong VWY., et al. (2012). Nat. Cell Biol. 14(4); 401-408).
